cGAS exacerbates Schistosoma japonicum infection in a STING-type I IFN-dependent and independent manner

Plos.org

by Le Liang, Yujuan Shen, Yuan Hu, Haipeng Liu, Jianping Cao

Schistosomiasis, which is caused by infection with Schistosoma spp., is characterized by granuloma and fibrosis in response to egg deposition. Pattern recognition receptors are important to sense invading Schistosoma, triggering an innate immune response, and subsequently shaping adaptive immunity. Cyclic GMP-AMP synthase (cGAS) was identified as a major cytosolic DNA sensor, which catalyzes the formation of cyclic GMP-AMP (cGAMP)... Читать дальше...

GDP polyribonucleotidyltransferase domain of vesicular stomatitis virus polymerase regulates leader-promoter escape and polyadenylation-coupled termination during stop-start transcription

Plos.org

by Minako Ogino, Todd J. Green, Tomoaki Ogino

The unconventional mRNA capping enzyme (GDP polyribonucleotidyltransferase, PRNTase) domain of the vesicular stomatitis virus (VSV) L protein possesses a dual-functional "priming-capping loop" that governs terminal de novo initiation for the leader RNA synthesis and capping of monocistronic mRNAs during the unique stop-start transcription cycle. Here, we investigated the roles of basic amino acid residues on a helix structure directly connected... Читать дальше...

Study of thin layer film evolution near bacterial cellulose membrane by Ag|AgCl electrodes in chamber with lower concentration

Plos.org

by Sławomir Grzegorczyn, Andrzej Ślęzak

We used the method of measuring potential difference between two Ag|AgCl electrodes immersed directly into electrolyte solution with lower concentration and at different distances from membrane. The bacterial cellulose membrane was placed in horizontal plane in the membrane system with configurations with higher NaCl concentration and density under (A) and over the membrane (B). In both configurations at the initial moment the voltage between electrodes amounted to zero. Читать дальше...

Comparing the impact of vaccination strategies on the spread of COVID-19, including a novel household-targeted vaccination strategy

Plos.org

by André Voigt, Stig Omholt, Eivind Almaas

With limited availability of vaccines, an efficient use of the limited supply of vaccines in order to achieve herd immunity will be an important tool to combat the wide-spread prevalence of COVID-19. Here, we compare a selection of strategies for vaccine distribution, including a novel targeted vaccination approach (EHR) that provides a noticeable increase in vaccine impact on disease spread compared to age-prioritized and random selection vaccination schemes. U.S. postdoctoral careers in life sciences, physical sciences and engineering: Government, industry, and academia

Plos.org

by Maya Denton, Maura Borrego, David B. Knight

Discussions about science and engineering postdoctoral researchers focus almost exclusively on academic postdocs and their chances of eventually securing tenure-track faculty positions. Further, biological sciences dominate policy research and published advice for new PhDs regarding postdoctoral employment. Our analysis uses the Survey of Earned Doctorates and Survey of Doctorate Recipients to understand employment implications for physical sciences... Читать дальше...

The complete chloroplast genome sequencing analysis revealed an unusual IRs reduction in three species of subfamily Zygophylloideae

Plos.org

by Xiaoyang Wang, Tashi Dorjee, Yiru Chen, Fei Gao, Yijun Zhou

Tetraena mongolica, Zygophyllum xanthoxylon, and Z. fabago are three typical dryland plants with important ecological values in subfamily Zygophylloideae of Zygophyllaceae. Studies on the chloroplast genomes of them are favorable for understanding the diversity and phylogeny of Zygophyllaceae. Here, we sequenced and assembled the whole chloroplast genomes of T. mongolica, Z. xanthoxylon, and Z. fabago, and performed comparative genomic and phylogenetic analysis. Читать дальше...

Biolayer interferometry for DNA-protein interactions

Plos.org

by John K. Barrows, Michael W. Van Dyke

Biolayer interferometry (BLI) is a widely utilized technique for determining macromolecular interaction dynamics in real time. Using changes in the interference pattern of white light reflected off a biosensor tip, BLI can determine binding parameters for protein-protein (e.g., antibody-substrate kinetics) or protein-small molecule (e.g., drug discovery) interactions. However, a less-appreciated application for BLI analysis is DNA-protein interactions. Читать дальше...

People’s perceptions of, willingness-to-take preventive remedies and their willingness-to-vaccinate during times of heightened health threats

Plos.org

by Angela Bearth, Anne Berthold, Michael Siegrist

Pandemics, such as the current SARS-CoV-2 pandemic, represents a health threat to humans worldwide. During times of heightened health risks, the public’s perceptions, and acceptance of evidence-based preventive measures, such as vaccines, is of high relevance. Moreover, people might seek other preventive remedies to protect themselves from getting infected (e.g., herbal remedies, nutritional supplements). A recent study on consumers’ preference...
