Williams, 34, did not start Wales’ opening two qualifiers – the 1-0 win over Slovakia in March and Saturday’s 2-1 loss in Croatia.
But Giggs has opted to call on the experience and leadership of the centre-back, who will win his 86th cap and move one ahead of the late Gary Speed into third place in the all-time appearance table.
Gareth Bale skippered Wales in Osijek but Williams will take over the armband for the big Group E showdown.
And Williams will be looking to put in a big display as his Toffees contract expires at the end of this month.
Wales chief Giggs revealed previously Williams has plenty of offers on the table with the likes of Aston Villa, Huddersfield and MLS duo DC United and New York City all keen.
Giggs has rung the changes after the draining conditions in Croatia where his side played in almost 30 degree heat.
Wales‘ all-time caps holder Chris Gunter is in line to win his 95th cap in place of Connor Roberts at right-back.
SunSport also understands Welsh Footballer of the Year David Brooks – the goalscorer in Osijek – and Ethan Ampadu, will both start after impressive cameos off the bench at the weekend.

Bournemouth ace Brooks has been carrying a knock and only trained two days before the Croatia game while Chelsea young gun Ampadu has been nursing a back complaint.
Matty Smith and Will Vaulks are expected to drop to the bench while Derby’s Tom Lawrence could also bring freshness to the side.
Wales need a positive result in the Hungarian capital as defeat would put them six points behind their rivals heading into the autumn.
