Paramedics and police had raced to a home at 7.30am amid fears children had been poisoned.
Two boys, aged 14 and 13, died after frantic attempts to resuscitate them.
Two girls, aged seven months and three years, and two younger boys — ten and 11 — were said to be doing well at Sheffield Children’s Hospital.
Police confirmed a woman of 34 and a man of 37 had been arrested on suspicion of murder.
Forensic officers were examining a three-bed semi in the Shiregreen neighbourhood of the city.
Police refused to confirm reports the victims were poisoned. Post-mortems were due to take place.
Neighbour Aaron Brunskill, 29, said there had been “about 20 police cars and four ambulances”.
He added: “Then the air ambulance came. They were just taking children away.”
Another neighbour said: “There’s a lot of talk that they may have been poisoned. It’s just too tragic to think about.”
